Expressions of MUC5AC and TNF-α in middle ear mucosa, and influence of anti-TNF-alpha; McAb treatment on rat model of acute otitis media with effusion

Abstract

Purpose: To investigate the effect of anti-TNF-α monoclonal antibody treatment (TNF-α McAb) on a rat model of acute otitis media with effusion (AOME).
Methods: Sprague Dawley rats of both sexes (n = 45) weighing 100 to 150 g were used for this study. They were randomly assigned to three groups: control group (n = 5), OME group (n = 20), and TNF-α McAb group (n = 20). The right ear of rats in OME and TNF-α McAb groups were treated as study ears, while their left ears served as control ears. Changes in tympanic membranes of right ears were closely monitored on days 1, 3, 7 and 14. Immunohistochemical staining and quantitative real-time polymerase chain reaction (qRT-PCR) were used to determine the expressions of MUC5AC and TNF-α in middle ear tissue sections.
Results: Treatment with anti-TNF-α McAb significantly increased the expression level of MUC5AC mRNA, and the expression peaked at day 3, and then declined time-dependently (p < 0.05). In OME group, the expression level of MUC5AC mRNA was significantly and time-dependently reduced (p < 0.05). Moreover, TNF-α mRNAs were significantly upregulated in OME and TNF-α McAb groups, relative to control group, and after peaking on day 3, declined time-dependently in both groups (p < 0.05).
Conclusion: Anti-TNF-α McAb promotes recovery from AOME.
